目的观察结肠癌组织血管、淋巴管内皮细胞podoplanin的表达,为研究癌淋巴管转移的分子机制提供形态学资料。方法根据不同病变阶段的手术病人切除的结肠癌标本进行HE染色和podoplanin的免疫组化染色,观察podoplanin在结肠癌组织中血管和淋巴管的表达情况。结果结肠癌组织血管内皮细胞podoplanin不染色,淋巴管内皮细胞podoplanin阳性表达,同时,随着临床分期的增高和淋巴结转移的发生,podoplanin的阳性表达率增高。结论podoplanin可选择性的表达在结肠癌组织淋巴管内皮上,能明确的区分血管及淋巴管成分,可作为一个淋巴管内皮特有的标志物。同时,癌组织内的淋巴管随着临床分期的增高和淋巴结转移的发生,podoplanin阳性表达率增高,这说明随着肿瘤的进展,结肠癌组织中的淋巴管数量随之增加,可能与结肠癌的淋巴转移有关。
Objective To observe the expression of podoplanin in vascular and lymphatic endothelial cells of colon cancer and provide morphological data for studying the molecular mechanism of lymphatic metastasis. Methods HE staining and immunohistochemistry of podoplanin were performed on colon cancer specimens from patients undergoing surgical resection at different stages of disease. The expression of podoplanin in blood vessels and lymphatic vessels in colon cancer tissues was observed. Results The expression of podoplanin in colorectal carcinoma was not stained and the expression of podoplanin in lymphatic endothelial cells was positive. At the same time, the positive expression of podoplanin increased with the increase of clinical stage and lymph node metastasis. Conclusion podoplanin can be selectively expressed on the lymphatic endothelium of colon cancer tissues, and can clearly differentiate the components of blood vessels and lymphatic vessels, which can be used as a unique marker of lymphatic endothelium. At the same time, with the increase of clinical stage and lymph node metastasis, the positive rate of podoplanin in lymphatic vessels in cancer tissues increased, which indicated that the number of lymphatic vessels in colon cancer tissues increased with the progression of the tumor, Lymphatic metastasis.
