目的观察加味蒌石汤对肾阴虚型多囊卵巢综合征(PCOS)模型小鼠卵巢功能的保护作用,并初步探究其作用机制。方法将54只小鼠随机分为正常对照组,模型组,达英-35组,加味蒌石汤高、中、低剂量组6组,每组9只。除正常对照组外,其余各组均采用脱氢表雄酮(DHEA)联合甲状腺素及利血平法制备肾阴虚型PCOS小鼠模型,各组给予相应药物进行预防性治疗。观察小鼠阴道脱落细胞和卵巢组织形态学改变,称量小鼠体质量及卵巢质量,计算卵巢指数,采用酶联免疫吸附法测定小鼠血清性激素[血清睾酮(T)、雌二醇(E2)、促卵泡刺激素(FSH)和黄体生成素(LH)]水平,抗苗勒氏管激素(AMH)、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)水平及cAMP/cGMP比值,从而评估加味蒌石汤对肾阴虚型PCOS模型小鼠的影响。结果与正常对照组比较,模型组小鼠体质量增量显著降低(P<0.05),动情周期紊乱,卵巢湿重及卵巢指数均显著升高(P<0.05),血清T,LH,cAMP,AMH水平及cAMP/cGMP比值均显著升高(P<0.01),血清E2,FSH,cGMP水平均显著降低(P<0.01);与模型组比较,加味蒌石汤中、高剂量组及达英-35组小鼠体质量增量均显著升高(P<0.05),卵巢湿重及卵巢指数均显著降低(P<0.05),各药物组动情周期逐渐恢复正常,血清T,LH,cAMP,AMH水平及cAMP/cGMP比值均显著降低,血清E2,FSH,cGMP水平均显著升高(P<0.05或P<0.01);各药物组之间比较,加味蒌石汤高、中剂量组及达英-35组对以上指标的改善作用显著优于加味蒌石汤低剂量组,差异均有统计意义(P<0.05)。结论加味蒌石汤能有效改善血清性激素T,E2,LH,FSH水平,且随药物剂量的增高效果越好;加味蒌石汤可通过调节AMH,cAMP水平和cAMP/cGMP比值改善PCOS模型小鼠肾阴虚症状,促进卵泡发育。
Objective To observe the protective effect of Jiawei Jieshi Decoction on the ovarian function in model mice with Kidney-Yin Deficiency Polycystic Ovary Syndrome (PCOS) and to explore its mechanism. Methods 54 mice were randomly divided into normal control group, model group, up-Ying-35 group, Jiawei Jieshi Tang high, medium and low dose group 6, each group 9. Except the normal control group, all the other groups were given DHEA combined with thyroxine and reserpine for the preparation of PCOS mouse model of kidney-yin deficiency, and the corresponding groups were given prophylactic treatment. The morphological changes of vaginal exfoliated cells and ovarian tissue of mice were observed. The body weight and ovary mass of mice were weighed. The ovarian index was calculated. Serum testosterone (T) and estradiol (E2) were measured by enzyme-linked immunosorbent assay , FSH and LH], anti-Müllerian hormone (AMH), cAMP, cGMP and cAMP / cGMP In order to evaluate the influence of Jiawei Jieshi decoction on mice with kidney-Yin-induced PCOS model. Results Compared with the normal control group, the increment of body weight in model group was significantly decreased (P <0.05), the estrous cycle was disordered, the ovarian wet weight and ovarian index were significantly increased (P <0.05) AMH, cAMP / cGMP, serum E2, FSH and cGMP were significantly decreased (P <0.01). Compared with model group, Jiawei Jieshi decoction medium and high dose group (P <0.05), wet weight of ovary and ovarian index (P <0.05), the estrous cycle of each drug group returned to normal gradually, serum T, LH, cAMP, AMH levels and cAMP / cGMP ratio were significantly decreased, serum E2, FSH, cGMP levels were significantly increased (P <0.05 or P <0.01); between the drug groups, The effect of improving the above indexes in Ying-35 group was significantly better than that in Jiawei Jieshi decoction group, the difference was statistically significant (P <0.05). Conclusions Jiawei Jieshi Decoction can effectively improve the levels of serum sex hormones T, E2, LH and FSH, and the better the effect with the increase of the dosage of drugs. Jiawei Jieshi decoction can improve the mice model of PCOS by adjusting AMH, cAMP level and cAMP / cGMP ratio Kidney Yin symptoms, promote follicular development.
