观察柴胡有效成分柴胡皂甙对原代培养贮脂细胞(FSC)激活及合成细胞外基质(ECM)的作用。结果:治疗组库普弗细胞条件培养基(TKCncm)、治疗CCl4损伤库普弗细胞条件培养基(TKCtcm)、TKCncm加肝细胞条件培养基(PCncm)组FSC内结蛋白阳性反应明显减弱,对细胞表型转化的形态学特征有一定改善,各治疗组FSC的DNA合成及3H-脯氨酸掺入量均呈不同程度降低,FSC内Ⅰ型胶原含量明显减少。说明柴胡皂甙对肝细胞具有保护作用,直接抑制FSC内DNA的合成,还可抑制FSC的激活,从而抑制FSC合成ECM的能力。
To observe the effect of saikosaponin, an effective component of Bupleurum chinense DC on the activation and synthesis of extracellular matrix (ECM) in primary cultured lipid-storage cells (FSC). RESULTS: The positive reaction of desmin in FSC was significantly decreased in the treatment group with Kupffer cell conditioned medium (TKCncm), CCl4 injury treated Kupffer cell conditioned medium (TKCtcm), TKCncm plus hepatocyte conditioned medium (PCncm) group. The morphological characteristics of cell phenotype transformation were improved. The DNA synthesis and 3H-proline incorporation of FSC in each treatment group decreased to some extent, and the content of type I collagen in FSC decreased significantly. It was demonstrated that saikosaponin has a protective effect on hepatocytes, directly inhibits DNA synthesis in FSC, and also inhibits the activation of FSC, thereby inhibiting the ability of FSC to synthesize ECM.
