目的观察固本咳喘颗粒(GKG)对慢性阻塞性肺疾病(COPD)大鼠可溶性白细胞介素-2受体(s IL-2R)表达的影响。方法用烟熏加气管内滴注脂多糖建立COPD大鼠模型,随机分为6组:正常组、模型组、3个剂量GKG组及固本咳喘片组。从造模的第3天开始,正常组和模型组灌服0.9%氯化钠10m L·kg-1,GKG组分别灌服3个剂量GKG混悬液(2.28,1.14,0.57 g·m L-1),固本咳喘片组灌服混悬液0.01 g·m L-1,灌胃1次/日,共28 d。灌胃结束第2天处死大鼠,留取标本。用双抗体夹心ABC-ELISA法检测大鼠s IL-2R。结果小剂量GKG组和固本咳喘片组能降低s IL-2R表达,与模型组比较差异有统计学意义(P<0.05)。结论 GKG可通过调控s IL-2R,从而提高COPD大鼠免疫功能及抗感染能力。
Objective To observe the effect of Gubeng Kechuan Granule (GKG) on the expression of sIL-2R in chronic obstructive pulmonary disease (COPD) rats. Methods Chronic obstructive pulmonary disease (COPD) rats were established by intragastric administration of lipopolysaccharide with smoke and gas. The rats were randomly divided into 6 groups: normal group, model group, 3 doses of GKG group and Guben Kechuan Tablet group. From the third day of modeling, normal group and model group were given 0.9% sodium chloride 10 m L · kg -1, GKG group were given three doses of GKG suspension (2.28,1.14,0.57 g · m L -1), Gubenchuchuan tablets group was fed with 0.01 g · m L-1 suspension, once a day for 28 days. The rats were sacrificed on the second day after gavage, and specimens were taken. Rat sIL-2R was detected by double antibody sandwich ABC-ELISA. Results Low-dose GKG and Guben Kechuan Tablet decreased the expression of IL-2R, which was significantly different from the model group (P <0.05). Conclusion GKG can improve the immune function and anti-infective ability of COPD rats through regulating s IL-2R.
