目的:研究肝细胞生长因子(HGF)对缺氧、缺糖诱导的星形胶质细胞(As)损伤及凋亡的影响。方法:以缺氧、缺糖来诱导原代培养的大鼠大脑皮层星形胶质细胞损伤。用不同终浓度(20~100ng/mL)的HGF处理星形胶质细胞,以乳酸脱氢酶(LDH)漏出率作为细胞损伤指标,四甲基偶氮唑盐(MTT)还原试验检测细胞活力,流式细胞术检测凋亡细胞,透射电镜观察细胞超微结构。结果:缺氧缺糖引起LDH漏出率增高,细胞活力下降,细胞凋亡率增高。HGF处理后,LDH漏出率降低,细胞活力增高,同时细胞凋亡率降低(P<0.05),最大效应剂量为60ng/mL。结论:HGF对星形胶质细胞缺氧缺糖损伤具有保护作用,能减少星形胶质细胞凋亡,并呈一定的剂量依赖关系。
Objective: To investigate the effects of hepatocyte growth factor (HGF) on hypoxia, glucose-induced astrocyte injury and apoptosis. Methods: Primary cultured rat cerebral cortical astrocytes were induced by hypoxia and glucose deprivation. Astrocytes were treated with different concentrations of HGF (20 ~ 100ng / mL), the leakage rate of lactate dehydrogenase (LDH) was taken as the indicator of cell damage, and the cell viability was measured by MTT reduction assay The apoptotic cells were detected by flow cytometry and the ultrastructure of cells were observed by transmission electron microscope. Results: LDH leakage rate increased, cell viability decreased and apoptosis rate increased after oxygen and glucose deprivation. HGF treatment, LDH leakage rate decreased, cell viability increased, while the apoptosis rate decreased (P <0.05), the maximum effective dose of 60ng / mL. Conclusion: HGF has a protective effect on hypoxia / hypoglycemic injury of astrocytes, which can reduce the apoptosis of astrocytes in a dose dependent manner.
