目的:探讨罗格列酮对人胚胎干细胞(hESCs)向脂肪细胞分化的作用。方法:收集常规IVF/ICSI-ET周期废弃胚胎,建立并鉴定hESCs系,扩增备用。根据基础成脂诱导分化培养基中是否添加10μmol/L罗格列酮分为实验组和对照组,采用油红-O染色测定细胞内脂滴、GPO-POD酶法测定细胞内甘油三酯和RT-PCR法测定脂肪细胞特异性基因过氧化物酶体增殖物激活受体γ(peroxisome proliferator-activated receptor gamma,PPARγ)mRNA鉴定脂肪细胞,并进行组间比较。结果:hESCs可定向诱导分化为脂肪细胞,罗格列酮刺激组较对照组甘油三酯含量高,差异具有显著性(P<0.05)。结论:PPARγ激动剂——罗格列酮对人胚胎干细胞向脂肪细胞诱导分化有促进作用,可显著提高诱导分化效率。
Objective: To investigate the effect of rosiglitazone on adipocyte differentiation of human embryonic stem cells (hESCs). Methods: The conventional IVF / ICSI-ET cycles were used to collect embryos, establish and identify hESCs lines and amplify them for later use. According to whether basal adipose-derived differentiation medium was added with 10μmol / L rosiglitazone into experimental group and control group, intracellular lipid droplets were measured by oil red-O staining and intracellular triglyceride was determined by GPO-POD enzymatic method The adipocytes were identified by RT-PCR assay of adipocyte-specific gene peroxisome proliferator-activated receptor gamma (PPARγ) mRNA, and compared between groups. Results: hESCs could be induced to differentiate into adipocytes. The triglyceride content of rosiglitazone - stimulated group was higher than that of control group (P <0.05). CONCLUSION: Rosiglitazone, a PPARγ agonist, can promote the differentiation of human embryonic stem cells into adipocytes and can significantly increase the efficiency of differentiation.
