目的筛选出在肝细胞癌组织中差异表达的长链非编码RNA(LncRNA),研究其表达水平与肝细胞癌临床病理特征的相关性。方法收集40对肝细胞癌与配对的正常肝组织标本,使用基因芯片结合权威数据库筛选出在肝细胞癌组织中差异表达的LncRNA,预试验确定作为研究目标的LncRNA,使用实时荧光定量PCR研究目标LncRNA在肝细胞癌组织中的表达情况,并分析其表达与肝细胞癌临床病理特征的相关性。结果选取基因芯片中差异表达倍数最明显的10个候选LncRNA进行预试验,确定Linc00152作为研究目标。85%(34/40)的肝细胞癌组织中Linc00152相对表达量高于配对正常肝组织。肝细胞癌低分化者的Linc00152低表达者比例高于中分化者(P<0.017),有血管侵犯者的Linc00152低表达者比例高于无血管侵犯者(P<0.05)。结论 LncRNA-Linc00152在肝细胞癌组织中表达升高,且和肝细胞癌的病理分级和血管侵犯相关。
Objective To screen long-chain non-coding RNA (LncRNA) differentially expressed in hepatocellular carcinoma (HCC) and to investigate its correlation with the clinicopathological features of hepatocellular carcinoma. Methods Forty pairs of hepatocellular carcinoma (HCC) and paired normal liver tissue samples were collected. LncRNAs differentially expressed in hepatocellular carcinoma tissues were screened by gene chip combined with authoritative database. The LncRNAs targeted by the experiment were determined in advance and the targets of real-time fluorescence quantitative PCR LncRNA expression in hepatocellular carcinoma, and analyze its correlation with clinicopathological features of hepatocellular carcinoma. Results Ten candidate LncRNAs with the most obvious differential expression in the gene chip were selected for pre-test, and Linc00152 was selected as the research target. The relative expression level of Linc00152 in 85% (34/40) hepatocellular carcinoma tissue was higher than that in normal liver tissue. The proportion of Linc00152 low expression in poorly differentiated hepatocellular carcinoma was higher than that in moderately differentiated hepatocellular carcinoma (P <0.017), and the low expression of Linc00152 in vascular infiltrated patients was higher than that in non-vascular invasion (P <0.05). Conclusion The expression of LncRNA-Linc00152 is up-regulated in hepatocellular carcinoma and is related to the pathological grade and vascular invasion of hepatocellular carcinoma.
