目的建立球花石斛离体快繁研究体系。方法利用球花石斛种子为外植体,采用种子培养方式开展繁育试验研究。结果球花石斛种子接种MS+香蕉泥13%的培养基上,可成功诱导其原球茎;MS+0.1mg/L NAA+1.5mg/L 6-BA+香蕉汁13%为球花石斛增殖的最佳培养基,增殖系数为7.9;无根苗生根的适宜培养基为1/2 MS+0.1mg/L NAA+2mg/L 6-BA+香蕉汁13%+土豆汁10%+活性炭2mg/L,生根率达到95%,平均根数为6.3条;组培养移栽到松树皮+水苔(3∶1)的栽培基质上,成活率为100%,且植株长势良好。结论寻找到球花石斛离体繁育研究中各阶段最佳培养基,建立了该植物的离体快繁体系。
Objective To establish a rapid propagation system of Dendrobium candidum in vitro. Methods The seeds of Dendrobium candidum were used as explants, and breeding experiments were conducted using seed culture methods. Results The seeds of D. officinale were inoculated with 13% medium of MS + banana puree, and the protocorm could be successfully induced. MS + 0.1 mg / L NAA + 1.5 mg / L 6-BA + banana juice 13% Medium with multiplication coefficient of 7.9; rooting medium without rooting was 1/2 MS + 0.1 mg / L NAA + 2 mg / L 6-BA + banana juice 13% + potato juice 10% + activated carbon 2 mg / Reaching 95% with an average of 6.3. The survival rate of the group was transplanted into the cultivation substrate of pine bark + moss (3: 1), and the plant survival rate was 100%. Conclusion The optimal culture medium in vitro of Dendrobium candidum was found, and the in vitro propagation system was established.
