为阐明在肺纤维化过程中肺成纤维细胞(LF)产生白介素-8(IL-8)的动态变化及作用,动态观察并对比博莱霉素(BLM)致肺纤维化大鼠支气管肺泡灌洗液(BALF)和LF培养上清中IL-8活性。结果:①LF上清中IL-8活性于第1天明显增高,第7天达高峰,随后降低,第28天降到正常。②BALF中IL-8活性也于第1天明显升高,但第3天达峰值,随后降低,第28天降到正常。③BALF和LF上清中IL-8活性之间以及二者与BALF中细胞总数及分数之间均有明显的正相关。证明LF在肺纤维化早期产生IL-8明显增多,是肺内IL-8活性的主要来源之一。BALF和LF上清中IL-8活性的水平反映了肺泡炎的强度和疾病的活动性。
To clarify the dynamic changes of interleukin-8 (IL-8) produced by lung fibroblasts (LFs) during pulmonary fibrosis, the effects of bleomycin (BLM) on bronchoalveolar lavage IL-8 activity in washings (BALF) and LF culture supernatants. Results: ① IL-8 activity in the supernatant of LFP increased significantly on the first day and peaked on the seventh day, then decreased and then decreased to normal on the 28th day. ② The activity of IL-8 in BALF was also significantly increased on the first day, but reached the peak on the third day, then decreased and dropped to normal on the 28th day. ③ There was a significant positive correlation between the activity of IL-8 in BALF and LF supernatant and the total number and fraction of cells in BALF. Proof of LF IL-8 in the early stage of pulmonary fibrosis significantly increased, is one of the main sources of IL-8 activity in the lung. Levels of IL-8 activity in BALF and LF supernatants reflect the intensity of alveolitis and disease activity.
