目的:为寻找一种简便、经济、灵敏度高而不需使用放射性同位素的生物活性检测方法对成纤维细胞生长因子(FGF)进行检测。方法:利用肝素对Balbc3T3 细胞株进行诱导,然后运用结晶紫比色法对其进行生物活性检测。结果:最大吸光度A值的一半为0 .315,所对应的aFGF浓度为24 pgml,而没有肝素诱导的最大A值一半为0.16 ,所对应的浓度为156pgml。结论:肝素的存在提高了结晶紫法的灵敏度。因此是一种简便、快捷而值得推广应用的FGF生物活性检测法。
OBJECTIVE: To search for a simple, economical and sensitive biological activity assay that does not require radioisotope detection of fibroblast growth factor (FGF). Methods: The Balbc3T3 cell line was induced by heparin, and its biological activity was detected by crystal violet colorimetry. Results: Half of the maximum absorbance A value is zero. 315, corresponding to a concentration of aFGF of 24 pg / ml without heparin-induced maximum A value of half 0.16, corresponding to a concentration of 156 pg / ml. Conclusion: The presence of heparin increases the sensitivity of the crystal violet method. Therefore, it is a simple, rapid and worth promoting the application of FGF biological activity detection method.
