目的:观察六味地黄丸延缓5/6肾切除大鼠肾组织纤维化的作用。方法:SD大鼠随机分成正常对照组、模型组及六味地黄丸组;采用5/6肾切除术诱导大鼠肾小球硬化模型,于第5~8周分批处死大鼠。PAS染色观察大鼠残肾组织形态学改变;免疫组织化学方法分别检测肾组织中CollagenⅣ(Col-Ⅳ)/MMP-2的表达。结果:六味地黄丸组细胞增生明显轻于模型组,毛细血管开放较好,肾小管结构基本正常。Ⅳ型胶原在模型组中有较强的表达,尤其是在肾小管中染色很深,而在六味地黄丸组无论是肾小管还是肾小球着色均较浅,仅有少量表达;六味地黄丸组MMP-2表达强烈,染色明显深于模型组。结论:六味地黄丸可能通过促进MMP-2的表达,对抗Ⅳ胶原的过度沉积,减轻硬化程度,达到保护肾脏,延缓肾功能衰竭进程的积极作用。
Objective: To observe the effect of Liuwei Dihuang Wan on delaying renal fibrosis in 5/6 nephrectomized rats. Methods: Sprague-Dawley rats were randomly divided into normal control group, model group and Liuwei Dihuang Pill group. Rat glomerular sclerosis model was induced by 5/6 nephrectomy. Rats were killed in batches at 5-8 weeks. PAS staining was used to observe the histomorphological changes of rat remnant kidneys. Immunohistochemistry was used to detect the expression of Collagen IV (Col-IV)/MMP-2 in renal tissues. Results: The cell proliferation of the Liuwei Dihuang Pill group was significantly lighter than that of the model group. Capillary opening was better, and the renal tubular structure was basically normal. Type IV collagen was strongly expressed in the model group, especially in the renal tubules. However, in the Liuwei Dihuang Wan group, both the renal tubules and the glomeruli were lightly colored, with only a small amount of expression; Liu Wei Di Huang Wan The expression of MMP-2 in the group was intense and the staining was significantly deeper than that in the model group. Conclusion: Liuwei Dihuang Pill may promote the expression of MMP-2, counteract the excessive deposition of collagen IV, reduce the degree of sclerosis, protect the kidney and delay the active process of renal failure.
