目的:研究双参通脉滴丸对大鼠脑缺血再灌注损伤脑组织梗死面积和神经细胞凋亡的影响及该影响是否存在剂量依赖性。方法:采用双侧颈总动脉阻断的方法建造大鼠脑缺血再灌注损伤模型。将大鼠随机分为假手术组、模型组、双参通脉滴丸(高、中、低剂量)组。采用TTC染色法测定脑组织梗死面积,末端脱氧核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL)检测脑组织凋亡神经细胞。结果:双参通脉滴丸(高、中、低剂量)组TTC染色法测定脑组织梗死面积与模型组比较差异显著,与假手术组无统计学意义;采用TUNEL法检测脑组织凋亡神经细胞数与模型组比较有差异,与假手术组相近。结论:双参通脉滴丸能减轻大鼠脑缺血再灌注损伤后脑组织梗死面积,抑制脑神经细胞的凋亡,并且其作用存在剂量依赖性。
Objective: To study the effects of Shuangshen Tongmai Dripping Pills on cerebral infarct size and neuronal apoptosis after cerebral ischemia-reperfusion injury in rats and whether the effect is dose-dependent. Methods: Bilateral common carotid artery occlusion was used to establish a rat model of cerebral ischemia-reperfusion injury. The rats were randomly divided into sham operation group, model group and Shuangshen Tongmai drop pills (high, medium and low dose) groups. TTC staining was used to determine the infarct size of brain tissue. Apoptotic neurons were detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL). Results: The TTC staining method of Shuangshen Tongmai dripping pill (high, medium and low dose) group showed significant difference compared with model group, and there was no significant difference between the two groups. The TUNEL method was used to detect the apoptosis of brain tissue The number of cells was significantly different from the model group, similar to the sham-operated group. Conclusion: Shuangshen Tongmai Dripping Pills can reduce cerebral infarct size and inhibit the apoptosis of cerebral neurons after cerebral ischemia-reperfusion injury in rats, and its effect is dose-dependent.
