目的:通过观察大黄虫丸对Lewis肺癌小鼠血管内皮生长因子(VEGF)、环氧化酶-2(COX-2)表达的影响,及其对微血管生成的作用,探索其抗肿瘤的可能机制。方法:造模后次日先随机分成5组,每组10只。根据预试结果确定高(H)、中(M)、低(S)剂量;同时设空白组和化疗组。d11处死小鼠,剖取完整肿块称重并计算抑瘤率。以免疫组化法检测VEGF、COX-2的表达,并用血管内皮细胞标记物CD34单克隆抗体标记行免疫组化染色,观察肿瘤微血管生成状况。结果:实验组高(H)、中(M)、低(L)和化疗组抑瘤率分别为29.70%、38.79%、17.58%、63.64%。免疫组化法表明大黄虫丸具有抑制VEGF、COX-2表达的作用,同时降低肿瘤微血管生成。结论:大黄虫丸对荷Lewis肺癌小鼠肿瘤具有明显的抑制作用,作用机制可能与抑制VEGF及COX-2表达,降低肿瘤微血管生成有关。
Objective: To observe the effect of Dahuang Zhechong Pill on the expression of vascular endothelial growth factor (VEGF) and cyclooxygenase-2 (COX-2) in mice with Lewis lung cancer and its effect on angiogenesis and to explore its possible anti-tumor effect mechanism. Methods: The next day after modeling were randomly divided into 5 groups of 10. According to the pre-test results to determine high (H), medium (M), low (S) dose; at the same time set the blank group and chemotherapy group. Mice were sacrificed at d11, intact lumps were weighed and tumor inhibition rates calculated. The expression of VEGF and COX-2 was detected by immunohistochemistry. The expression of VEGF-C was detected by immunohistochemical staining with CD34 monoclonal antibody. The angiogenesis was observed. Results: The inhibitory rates of H, M, L and chemotherapy in the experimental group were 29.70%, 38.79%, 17.58% and 63.64%, respectively. Immunohistochemistry showed that Dahuang Shechong Pill inhibited the expression of VEGF and COX-2, and reduced the tumor angiogenesis. Conclusion: Rhubarb Zhechong Pills can obviously inhibit the tumor of mice bearing Lewis lung carcinoma and the mechanism may be related to inhibiting the expression of VEGF and COX-2 and decreasing the tumor angiogenesis.
