为检测新的凋亡相关基因PDCD5在狼疮性肾炎患儿肾组织中的表达 ,分析其与肾组织细胞凋亡的关系 ,探讨PDCD5在狼疮性肾炎发病中的作用 ,用免疫组化及图象分析的方法 ,检测19例狼疮性肾炎患儿肾组织及9例对照肾组织中PDCD5的表达 ;同时16/19例狼疮性肾炎和6/9例对照肾组织标本运用原位末端标记法(TUNEL)检测肾组织中的细胞凋亡。结果显示 :①在肾组织中可检测到PDCD5的表达 ,肾小管的表达量(0.20±0.02)强于肾小球的表达量 (0.09±0.01,P<0.05) ;②在狼疮性肾炎患儿肾组织中 ,PDCD5在肾小管的表达量 (0.13±0.01)强于肾小球的表达量 (0.07±0.01 ,P<0.05) ,PDCD5在肾小球的表达与对照组相比差异无显著性 ,在肾小管的表达明显低于对照组(P<0.05) ;③狼疮性肾炎患儿肾组织中肾小球、肾小管的凋亡细胞数均明显高于对照组(P<0.05)。本研究用免疫组化的方法首次检测了新的凋亡相关基因PDCD5在肾组织中的表达及分布 ;并发现在狼疮性肾炎患儿肾组织中PDCD5在肾小球的表达与对照组相比差异无显著性 ;在肾小管 ,虽然凋亡细胞数增加 ,但PDCD5的表达却下降。
To detect the expression of PDCD5, a novel apoptosis-related gene, in renal tissue of children with lupus nephritis and analyze the relationship between PDCD5 and apoptosis in renal tissue and to explore the role of PDCD5 in the pathogenesis of lupus nephritis. Immunohistochemistry and imaging 19 cases of lupus nephritis and 9 cases of control kidney were examined for the expression of PDCD5. Meanwhile, 16/19 cases of lupus nephritis and 6/9 cases of control renal tissues were detected by TUNEL ) To detect apoptosis in renal tissue. The results showed that: (1) The expression of PDCD5 was detected in renal tissues, the expression of tubulointerstitial (0.20 ± 0.02) was stronger than that of glomeruli (0.09 ± 0.01, P <0.05); ② In children with lupus nephritis The expression of PDCD5 in renal tubules (0.13 ± 0.01) was significantly higher than that in glomeruli (0.07 ± 0.01, P <0.05). There was no significant difference in the expression of PDCD5 in glomeruli between the two groups (P <0.05). ③The number of apoptotic cells in renal glomerulus and tubule in lupus nephritis patients was significantly higher than that in control group (P <0.05). In this study, we first detected the expression and distribution of PDCD5 in renal tissues by immunohistochemistry. We found that the expression of PDCD5 in glomerulus in renal tissues of patients with lupus nephritis was significantly lower than that of the control group No significant; in the renal tubules, although the number of apoptotic cells increased, PDCD5 expression decreased.
