在生物学X射线显微分析中,我们主要利用的是能谱仪提供的元素谱线。但这些谱线往往并不与样品中所含元素相一致。这种不一致统称做人工伪差。显然,分析这些伪差的来源及防止方法,对提高定性分析的准确性和定量分析的精确性是有益的。人工伪差主要有三种来源:1.来自样品制备过程中产生的伪差包括在固定、脱水、包埋、捞片、染色等过程发生的元素流失、再分配、引进外来元素等。关于这种伪差文献很多,这里就不讨论了。
In biological X-ray microanalysis, we mainly use the elemental spectrum provided by the spectrometer. However, these lines are often not consistent with the elements contained in the sample. This inconsistency collectively referred to as artificial artifacts. Obviously, analyzing the origin of these artifacts and preventing them is helpful to improve the accuracy of qualitative analysis and the accuracy of quantitative analysis. There are three main sources of artificial artifacts: 1. The artifacts arising from the sample preparation process include loss of elements, redistribution, introduction of foreign elements, etc. during immobilization, dehydration, embedding, skimming, staining and the like. A lot of literature on this artifact, not discussed here.
