目的以提取天然香菇菌多糖和茶叶多糖混合物为佐剂,探讨混合型植物多糖对血吸虫疫苗的保护性免疫增效作用。方法构建日本血吸虫重组质粒pVIVO2-IL12-Sj23。BALB/c小鼠随机分为3组,每组10只。A组每鼠股四头肌注射生理盐水100μl为对照,B组每鼠注射pVIVO2-IL12-Sj23 100μg,C组每鼠注射pVIVO2-IL12-Sj23 100μg加等量多糖混合物。小鼠接种后第4周以日本血吸虫尾蚴40±2条经皮肤攻击感染,感染后6周剖杀,计算减虫率及每克肝组织减卵率,同时用ELISA测定血清中特异性抗体水平。结果C组减虫率和每克肝组织减卵率分别为64.3%和79.9%,B组则分别为45.5%和58.3%,B、C两组的差异有统计学意义(P<0.05)。B、C两组IgG水平均较A组显著升高(P<0.05),但B、C两组IgG抗体水平的差异无统计学意义(P>0.05)。结论以天然香菇菌多糖和茶叶多糖混合物为佐剂,对日本血吸虫重组质粒pVIVO2-IL12-Sj23的免疫效果有一定的增效作用。
OBJECTIVE To extract a mixture of polysaccharides from natural lentinus edodes and tea polysaccharide as adjuvants, and to explore the protective immune synergistic effect of mixed plant polysaccharides on schistosoma vaccine. Methods To construct Schistosoma japonicum recombinant plasmid pVIVO2-IL12-Sj23. BALB / c mice were randomly divided into 3 groups of 10. A group of quadriceps injected saline 100μl as a control, B group injected with pVIVO2-IL12-Sj23 100μg, C group injected with pVIVO2-IL12-Sj23 100μg plus the same amount of polysaccharide mixture. Four weeks after inoculation, the mice were infected with 40 ± 2 schistosoma japonicum cercariae by skin attack and killed 6 weeks after infection. The worm reduction rate and the rate of egg reduction per gram of liver tissue were calculated. The level of serum specific antibody . Results The worm reduction rate and the rate of egg reduction per gram of liver were 64.3% and 79.9% in group C and 45.5% and 58.3% in group B, respectively. There was significant difference between group B and C (P <0.05). The levels of IgG in group B and group C were significantly higher than those in group A (P <0.05), but there was no significant difference in the level of IgG between groups B and C (P> 0.05). Conclusion The natural lentinan polysaccharide and tea polysaccharide mixture as adjuvant on the immune effect of recombinant plasmid pVIVO2-IL12-Sj23 Schistosoma japonicum has some synergies.
