目的研究工布乌头提取物对果蝇肠道免疫功能的影响。方法用标准的玉米粉酵母培养基饲养的果蝇作为对照组,标准培养基加入1.0%工布乌头提取物饲养的果蝇作为实验组,利用十二烷基硫酸钠(SDS)、葡聚糖硫酸钠(DSS)、过氧化氢(H2O2)和百草枯(PQ)等诱导肠道损伤,通过比较果蝇生存率、肠道上皮细胞凋亡情况、活性氧(ROS)量、肠道干细胞数量及肠道形态变化等,探讨工布乌头提取物对果蝇肠道的保护作用。结果 1.0%工布乌头提取物可以提高SDS、DSS、H2O2和PQ等致炎因子诱导的果蝇生存率;降低由SDS诱导的果蝇肠道细胞凋亡数量和肠道细胞内ROS量,减少肠道上皮细胞过度增殖,减少黑色素瘤的产生,并改善肠道形态。结论工布乌头提取物对致炎因子诱导的肠道损伤具有较好的保护作用。
Objective To study the effect of Aconitum kusnezoffii extract on intestinal immune function in Drosophila melanogaster. Methods Drosophila fed with standard cornmeal yeast culture medium was used as a control group. Drosophila fed with 1.0% Aconitum kusnezoffii extract was used as the experimental group. Sodium dodecyl sulfate (SDS) Intestinal injury was induced by DSS, H2O2 and paraquat (PQ). The survival rate of Drosophila, intestinal epithelial cell apoptosis, reactive oxygen species (ROS), gut stem cells Quantity and intestinal morphological changes, to explore the protective effect of Aconitum kusnezoffii extract on intestinal flies. Results 1.0% Aconitum kusnezoffii extract could increase the survival rate of Drosophila induced by inflammatory cytokines such as SDS, DSS, H2O2 and PQ, reduce the number of intestinal apoptosis induced by SDS and the amount of ROS in intestinal cells, Reduce the proliferation of intestinal epithelial cells, reduce the production of melanoma, and improve intestinal morphology. Conclusion Aconitum kusnezoffii extract has a good protective effect on inflammatory injury induced by intestinal tract.
