目的:探讨1,25二羟维生素D3[1,25(OH)2D3]与顺铂(DDP)联合抑制人子宫内膜癌HEC-1-A细胞的增殖,测定二者作用下S期激酶相关蛋白(Skp2)、抑癌基因p27蛋白的表达。方法:用CCK-8法测定1,25(OH)2D3、DDP及二者联合处理后细胞增殖情况,用流式细胞仪分析细胞周期,用W estern blot检测Skp2及p27蛋白表达的变化。结果:与对照组相比,1,25(OH)2D3、DDP作用下细胞吸光度(OD)值减少,细胞增殖受到抑制(P<0.01),二者联用时此作用明显增强;1,25(OH)2D3、DDP均能增加G0/G1期细胞比例,降低S、G2/M期细胞比例,从而降低细胞增殖指数(P<0.01);二者作用下Skp2表达减少,p27蛋白表达增多。结论:1,25(OH)2D3有增强DDP抗肿瘤的作用,二者均可能通过下调Skp2、上调p27蛋白表达,抑制癌细胞增殖。
AIM: To investigate the inhibitory effects of 1,25-dihydroxyvitamin D3 (1,25 (OH) 2D3] and cisplatin (DDP) on the proliferation of human endometrial carcinoma HEC-1-A cells. Protein (Skp2), tumor suppressor gene p27 protein expression. Methods: The proliferation of 1,25 (OH) 2D3, DDP and their combination were measured by CCK-8 assay. The cell cycle was analyzed by flow cytometry. The expression of Skp2 and p27 protein was detected by Western blot. Results: Compared with the control group, the cell viability (OD) of 1,25 (OH) 2D3 and DDP decreased and the cell proliferation was inhibited (P <0.01) OH) 2D3 and DDP both increased the proportion of cells in G0 / G1 phase and decreased the proportion of cells in S and G2 / M phases, thereby decreasing the cell proliferation index (P <0.01). Conclusion: 1,25 (OH) 2D3 can enhance the antitumor effect of DDP. Both of them may down-regulate Skp2, up-regulate the expression of p27 protein and inhibit the proliferation of cancer cells.
