目的:探索L-缬氨酸(L-Val)缺乏对大鼠胃癌与对照肌组织及体外培养胃癌细胞与成纤维细胞蛋白合成与免疫状态的影响。 方法:用[1-13C]Leu 示踪标记的“液闪法”测定大鼠胃癌与肌组织蛋白质合成率、用3 H-Leu 掺入法测定体外培养胃癌细胞与成纤维细胞蛋白质合成。 结果:14 只大鼠胃癌组织经无L-Val输液组(L-Valdeprivated solution,DS)作用后蛋白合成率显著低于L-Val平衡输液组(L-Valbalanced solution,BSn= 10),P<0.01;对照肌组织经BS、DS作用后蛋白合成率差异不显著。MKN45 胃癌细胞株BS组蛋白合成率显著高于DS组(P< 0.05),NIH3T3成纤维细胞株这两组蛋白合成差异不显著。DS作用过的负胃癌大鼠血清白蛋白、转铁蛋白、前白蛋白及C3含量与BS组比较差异不显著。 结论:配制一种既阻碍癌肿生长,又不引发宿主负氮平衡和免疫力下降的无L-Val的氨基酸输液治疗胃癌可能有很大的临床应用价值
OBJECTIVE: To investigate the effects of L-Val deficiency on protein synthesis and immune status of gastric cancer and control muscle tissues and cultured gastric cancer cells and fibroblasts in vitro. Methods: The rate of protein synthesis in gastric cancer and muscle tissue was measured by “liquid scintillation method” with [1-13C]Leu tracer, and the protein synthesis of gastric cancer cells and fibroblasts was measured by 3 H-Leu incorporation. RESULTS: The protein synthesis rate of 14 gastric cancer tissues after L-Valdeprivated solution (DS) was significantly lower than that of L-Val balanced solution (BSn=10), P< 0.01; There was no significant difference in the protein synthesis rate between the control muscle tissue and BS and DS. The rate of protein synthesis in the MKN45 gastric cancer cell line BS group was significantly higher than that in the DS group (P<0.05). There was no significant difference in the protein synthesis of the NIH3T3 fibroblast cell line between the two groups. There were no significant differences in serum albumin, transferrin, prealbumin, and C3 levels in negative stomach cancer rats with DS. Conclusion: The preparation of a kind of amino acid infusion without L-Val that can inhibit the growth of cancer without causing the negative balance of nitrogen in the host and the decrease of immunity may have great clinical value in the treatment of gastric cancer.
