目的:观察体外培养的大鼠肾小球系膜细胞在内毒素(LPS)诱导下IL┐6的产生及其mRNA表达。方法:按经典方法培养肾小球系膜细胞(MC)、转种培养后分为二组,诱导组加LPS10mg/L,对照组不加,用分子生物学和免疫学方法进行检测。结果:IL┐6在正常MC中分泌量极微,其mRNA也为低水平表达,经LPS诱导后二者均明显增加。IL┐6蛋白的产生6~12h达高峰,比对照组增加3~5倍,IL┐6mRNA的表达3~6h达高峰,较对照组增加2~3倍。结论:LPS可诱发系膜细胞IL┐6水平增高,可能作为炎症介质参与肾脏疾病的发病,在系膜增殖性肾炎发病中起一定作用
OBJECTIVE: To observe the IL-6 production and its mRNA expression induced by endotoxin (LPS) in cultured rat glomerular mesangial cells. Methods: The mesangial cells (MC) were cultured according to the classical method and divided into two groups after induction of culture. The induced group was treated with LPS 10 mg / L, while the control group was not added. The molecular biology and immunological methods were used to detect the mesangial cells. Results: IL-6 secreted in normal MC was extremely low, and its mRNA was also low-level expression, both of which were significantly increased after induced by LPS. The production of IL-6 protein peaked at 6 to 12 hours and increased 3 to 5 times than that in control group. The expression of IL-6 mRNA peaked at 3 to 6 hours, which was 2 to 3 times higher than that in control group. CONCLUSION: LPS can induce an elevated level of IL-6 in mesangial cells, which may play an important role in the pathogenesis of mesangial proliferative glomerulonephritis as an inflammatory mediator
