目的为评价红曲治疗大鼠骨折的疗效并探讨其作用机理。方法取12周龄的Wistar大鼠90只,手术法完成大鼠骨折模型后,随机分为3组,分别为治疗组(红曲)、阳性对照组(伤科接骨片)和空白对照组(生理盐水);分组饲养给药2、4、6周,分别行摄X线片、BMP-2免疫组化观察、组织学观察。结果术后第4周X片显示治疗组骨折线趋于消失,骨折端有大量连接性骨痂阴影,基本上达到骨愈合。术后第6周治疗组骨折线基本消失,可见髓腔再通;对照组骨折线趋于消失,骨折端及周围有大量连接线骨痂阴影,空白组中有出现断端硬化不愈合现象。治疗组在胞浆中有BMP-2的棕色深染,染色程度明显高于空白对照组(P<0.05)。结论红曲明显促进了大鼠的骨痂形成,提高BMP-2阳性细胞率,促进间充质细胞分泌BMP-2,并促使其向成骨细胞转化从而加速骨形成,从而具有促进骨折愈合作用。
Objective To evaluate the efficacy of red yeast in the treatment of fractures in rats and to explore its mechanism of action. Methods Ninety Wistar rats (12 weeks old) were used to complete the fracture model by surgery. The fractures were randomly divided into 3 groups: treatment group (red yeast), positive control group (Jianke Jiegu tablets) and blank control group ( Normal saline); group fed for 2, 4, and 6 weeks, respectively, taking X-ray film, BMP-2 immunohistochemical observation, histological observation. RESULTS: The X-ray showed that the fracture line of the treatment group disappeared at the 4th week postoperatively. There was a large number of shadows of connective osteophytes at the fracture end, which basically reached bone healing. At the 6th week after operation, the fracture line of the treatment group basically disappeared and the medullary cavity was recanalized; the fracture line of the control group tended to disappear, and there were a large number of shadows of the connective bone at the end of the fracture. There was a phenomenon of non-union of stump sclerosis in the blank group. In the treatment group, BMP-2 was stained brown in the cytoplasm, and the degree of staining was significantly higher than that in the control group (P<0.05). Conclusion Monascus obviously promotes the formation of callus in rats, increases the rate of BMP-2 positive cells, promotes the secretion of BMP-2 by mesenchymal cells, and promotes the conversion of osteoblasts to osteoblasts to accelerate bone formation, thus promoting the healing of fractures. .
