目的:观察腹腔注射不同剂量长托宁对大鼠空间学习、记忆、大鼠海马组织Bax/Bcl-2蛋白表达和乙酰胆碱酯酶(AChE)活性的影响。方法:9月龄SD大鼠48只,随机分为4组(n=12)。对照组(C组):腹腔注射生理盐水2ml;长托宁0.64mg/kg(P1)组;长托宁1.28mg/kg(P2)组和长托宁2.56mg/kg(P3)组。停药1d后进行Morris水迷宫行为测试。测试结束后立即处死大鼠,取海马组织,进行免疫组织化学染色,测定海马Bax/Bcl-2蛋白的表达;用比色法进行AChE活性测定。结果:水迷宫测试结果:P3组和P2组大鼠大鼠的潜伏期(2-3d)和游泳距离均明显长于P1组和C组(P<0.05)。Bax蛋白、Bcl-2蛋白在P3组和P2组海马内表达及二者的比值均显著高于P1组和C组(P<0.05)。结论:腹腔重复注射较大剂量长托宁对大鼠的空间学习和记忆能力有明显损害,可能与其影响大鼠海马神经元的凋亡和胆碱能系统有关。
OBJECTIVE: To observe the effect of intraperitoneal injection of penehyclidine hydrochloride on spatial learning and memory, Bax / Bcl-2 protein expression and acetylcholinesterase (AChE) activity in hippocampus of rats. Methods: Forty-eight SD rats aged 9 months were randomly divided into 4 groups (n = 12). Control group (group C): intraperitoneal injection of saline 2ml; penehyclidine 0.64mg / kg (P1) group; penehyclidine 1.28mg / kg (P2) group and penehyclidine 2.56mg / kg (P3) group. Morris water maze test was performed 1 day after drug withdrawal. Rats were sacrificed immediately after the test, hippocampal tissues were taken for immunohistochemical staining to determine the expression of Bax / Bcl-2 protein in hippocampus. AChE activity was measured by colorimetric assay. Results: The results of water maze test showed that latency and swimming distance of rats in P3 and P2 groups were significantly longer than those in P1 and C groups (P <0.05). The expressions of Bax and Bcl-2 in P3 and P2 hippocampus were significantly higher than those in P1 and C groups (P <0.05). CONCLUSION: Greater intraperitoneal injection of penehyclidine hydrochloride significantly impairs the learning and memory ability of rats, which may be related to its effect on apoptosis and cholinergic system in hippocampal neurons of rats.
