目的检测Hedgehog信号通路中SHH蛋白及其下游转录因子GLI1蛋白在人Peutz-Jeghers综合征(PJS)息肉中的表达,探讨其在PJS发生及恶变中的作用。方法采用免疫组织化学方法检测PJS息肉(20例)、正常肠黏膜组织(20例)、大肠腺瘤性息肉(25例)、大肠腺癌(25例)组织中SHH和GLI1蛋白的表达和分布,并比较表达程度的差异。结果免疫组织化学检测SHH蛋白和GLI1蛋白的阳性着色主要分布于胞膜和胞质,腺癌中GLI1蛋白的胞质和核染色增加。SHH蛋白和GLI1蛋白在正常肠黏膜组织、PJS息肉组织、腺瘤组织及腺癌组织中的阳性表达率和表达强度依次增高,差异具有统计学意义(Kruskal-Wallis H test,P<0.001)。SHH与GLI1的蛋白表达在PJS中经Spearman等级相关分析呈正相关(rp=0.503,P=0.024)。结论 SHH/GLI1在正常肠黏膜组织、PJS息肉组织、腺瘤组织及腺癌组织中均有表达,且表达水平依次增高,提示SHH-GLI1信号通路可能与PJS息肉的发生及恶变有关。
Objective To detect the expression of SHH protein and its downstream transcription factor GLI1 protein in human Peutz-Jeghers syndrome (PJS) polyps in Hedgehog signaling pathway, and to explore its role in the pathogenesis of PJS and malignant transformation. Methods The expression and distribution of SHH and GLI1 in PJS polyps (20 cases), normal intestinal mucosa (20 cases), adenomatous polyps (25 cases) and adenocarcinoma of the colon (25 cases) were detected by immunohistochemistry. , And compare the difference of expression level. Results The positive staining of SHH protein and GLI1 protein detected by immunohistochemistry was mainly distributed in membrane and cytoplasm. The cytoplasm and nuclear staining of GLI1 protein in adenocarcinoma increased. The positive expression rate and expression intensity of SHH protein and GLI1 protein in normal intestinal mucosa tissue, PJS polyp tissue, adenoma tissue and adenocarcinoma tissue were increased in turn, with statistical significance (Kruskal-Wallis H test, P <0.001). The protein expression of SHH and GLI1 was positively correlated with PJS by Spearman rank correlation analysis (rp = 0.503, P = 0.024). Conclusions SHH / GLI1 is expressed in normal intestinal mucosa, PJS polyp, adenoma and adenocarcinoma, and the expression levels of SHH / GLI1 are successively increased, suggesting that SHH-GLI1 signaling may be related to the occurrence and malignant transformation of PJS polyps.
