目的研究甲状腺乳头状癌中叉头框蛋白M1(Fox M1)基因的表达以及其对丝裂原活化蛋白激酶(MAPK)信号通路中的重要基因Ras及CDK1的影响,以探讨Fox M1在甲状腺乳头状癌中是否通过MAPK通路发挥作用。方法用h Fox M1-RNA干扰处理甲状腺乳头状癌TPC-1细胞,抑制甲状腺乳头状癌TPC-1细胞中Fox M1基因的表达,使用实时荧光定量PCR法检测处理后的TPC-1癌细胞和未经处理的TPC-1癌细胞中Ras和CDK1基因的表达水平变化,观察甲状腺乳头状癌TPC-1细胞中Fox M1的表达对Ras和CDK1的影响。结果与未经处理的甲状腺乳头状癌TPC-1细胞相比,使用h Fox M1-RNA干扰处理后的TPC-1细胞中Fox M1 m RNA表达明显降低(0.452 9比1.005 0,t=24.692 9,P<0.01),Ras m RNA的表达相应升高(1.319 0比1.001 2,t=14.218 5,P<0.01),而CDK1 m RNA的表达相应降低(0.767 5比1.008 1,t=10.763 4,P<0.01)。结论在甲状腺乳头状癌TPC-1细胞中Fox M1基因表达水平可对Ras和CDK1的表达产生影响,其在甲状腺乳头状癌TPC-1细胞中的作用机制可能与MAPK信号通路有关。
Objective To investigate the expression of Fox M1 gene in papillary thyroid carcinoma and its effect on the expression of Ras and CDK1 in mitogen-activated protein kinase (MAPK) signaling pathway in order to explore the role of Fox M1 in thyroid papillary Whether or not MAPK pathway plays a role in cancer. Methods The expression of Fox M1 gene in papillary thyroid carcinoma TPC-1 cells was inhibited by h Fox M1-RNA interference. The expression of Fox M1 gene in papillary thyroid carcinoma TPC-1 cells was detected by real-time fluorescence quantitative PCR. The changes of Ras and CDK1 gene expression in untreated TPC-1 cancer cells were observed and the effect of Fox M1 expression on Ras and CDK1 in papillary thyroid carcinoma TPC-1 cells was observed. Results Fox M1 m RNA expression was significantly decreased in TPC-1 cells treated with h Fox M1-RNA compared with untreated thyroid papillary carcinoma TPC-1 cells (0.452 9 vs 1.005 0, t = 24.692 9 , P <0.01), the expression of Ras m RNA increased accordingly (1.3190 vs 1.001 2, t = 14.218 5, P <0.01), while the expression of CDK1 m RNA decreased accordingly (0.767 5 vs 1.008 1, t = 10.763 4 , P <0.01). Conclusion The expression of Fox M1 gene in TPC-1 cells of thyroid papillary carcinoma may affect the expression of Ras and CDK1, which may be related to the MAPK signaling pathway in papillary thyroid carcinoma TPC-1 cells.
