通过检测MKN28人胃癌细胞株~3H-胸腺嘧啶核苷掺人情况,在体外实验研究加温和抗癌药物顺氨氯铂、5-氟脲嘧啶之间的协同作用。结果表明(1)顺氨氯铂的细胞杀伤作用具有温度-药物浓度依赖性;(2)5-氟脲嘧啶合并加温未见有显著协同作用,反见削弱其抗肿瘤效应;(3)单纯加温与合并应用中等剂量的顺氨氯铂或5-氟脲嘧啶的比较中发现,效果最佳的为加温加顺氨氯铂,其次是单纯加温;(4)加温加顺氨氯铂、5-氟脲嘧啶组合治疗效果最佳。为临床应用治疗胃癌的腹腔广泛转移提供了理论依据。
By examining the incorporation of ~3H-thymidine in the MKN28 human gastric cancer cell line, the synergy between the warming and anticancer drug cisplatin and 5-fluorouracil was studied in vitro. The results showed that (1) the cell killing effect of cisplatin was temperature-drug concentration-dependent; (2) 5-fluorouracil combined with warming had no significant synergistic effect, but the anti-tumor effect was weakened; (3) The comparison between simple heating and the application of moderate doses of cisplatin or 5-fluorouracil found that the best effect was warming plus cisplatin, followed by simple warming; (4) heating plus cisplatin, The combination of 5-fluorouracil has the best effect. It provides a theoretical basis for the clinical application of the treatment of gastric cancer with extensive abdominal metastasis.
