目的 研究肿瘤抗原激活的树突状细胞 (DC)在肝癌病人体内诱导的抗肿瘤免疫能否抑制肝癌术后复发和转移。方法 分离肝癌根治性切除术后病人外周血DC ,以人肝癌细胞系HepG2细胞粗提物激活DC ,以粒 (巨噬 )细胞集落刺激因子 (GM CSF)和白介素 4(IL 4)联合刺激DC ,DC回输病人 ,检测肝癌病人外周血AFPmRNA。结果 在AFPmRNA阳性组中 ,治疗组 11例中仅 1例AFPmRNA仍为阳性 ,而对照组 11例中 9例AFPmRNA仍为阳性 ;在AFPmRNA阴性组中 ,治疗组 13例AFPmRNA仍全部为阴性 ,而对照组 13例中 4例AFPmRNA为阴性。结论 肿瘤抗原激活的DC在肝癌病人体内诱导的抗肿瘤免疫能够抑制术后肝癌复发和转移。
Objective To investigate whether anti-tumor immunity induced by tumor antigen-activated dendritic cells (DC) in liver cancer patients can inhibit the recurrence and metastasis of liver cancer. METHODS: Peripheral blood DCs were isolated from patients undergoing radical resection of hepatocellular carcinoma. DCs were stimulated with human hepatocellular carcinoma cell line HepG2 cells and stimulated with GM CSF and IL-4. , DC reinfusion of patients, detection of liver cancer patients peripheral blood AFPmRNA. Results In the AFP mRNA positive group, only 1 AFP mRNA was still positive in 11 patients in the treatment group, while 9 AFP mRNAs were still positive in the 11 patients in the control group. In the AFP mRNA negative group, 13 AFP mRNAs were still negative in the treatment group. In the control group, 4 out of 13 patients were negative for AFP mRNA. Conclusion Anti-tumor immunity induced by tumor antigen-activated DC in liver cancer patients can inhibit the recurrence and metastasis of postoperative liver cancer.
