目的探讨植物血球凝集素(PHA)在体外定向诱导人脐血单个核细胞(UBMNC)分化为内皮细胞的可行性。方法分离人UBMNC加入PHA体外诱导培养1~8d,在倒置显微镜下动态观察细胞的生长分化,并以流式细胞术(FCM)检测培养的细胞中Von Willebrand因子(vWF)的表达,在电镜下观察内皮细胞特异性的Weibel-Palade(W-P)小体。结果在PHA的诱导下,UBMNC可分化为典型的梭形内皮细胞。FCM检测诱导分化效率达60%以上;电镜观察发现,在胞质中含有内皮细胞特异性的W-P小体。结论UBMNC在PHA的诱导下,可定向分化为内皮细胞,为用脐血治疗缺血性疾病的可能性提供了实验依据。
Objective To investigate the feasibility of using human hemagglutinin (PHA) to differentiate human umbilical cord blood mononuclear cells (UBMNC) into endothelial cells in vitro. Methods Human UBMNC was isolated and cultured in PHA for 1 ~ 8 days. The growth and differentiation of cells were observed under an inverted microscope. The expression of von Willebrand factor (vWF) in cultured cells was detected by flow cytometry (FCM) Endothelial-specific Weibel-Palade (WP) bodies were observed. Results Under the induction of PHA, UBMNC can differentiate into typical spindle-shaped endothelial cells. FCM detection of induction of differentiation efficiency of more than 60%; electron microscopy showed that in the cytoplasm contains endothelial cell-specific W-P bodies. Conclusion UBMNC can be induced to differentiate into endothelial cells under the induction of PHA, which provides experimental evidence for the possibility of treating ischemic diseases with cord blood.
