目的:揭示开花对铁皮石斛多糖质量分数及单糖组成的影响,为优质铁皮石斛培育提供依据。方法:采集铁皮石斛开花前样品、开花结束时经人工摘除花蕾与未摘花蕾的样品,采用苯酚-硫酸法测定铁皮石斛中多糖质量分数,柱前衍生HPLC检测多糖中的单糖组分。结果:摘花蕾样品与未摘花蕾样品比较,总多糖及多糖中的甘露糖、半乳糖醛酸、葡萄糖绝对量分别高30.35%,26.44%,20.78%,57.80%,木糖,阿拉伯糖分别低28.33%,24.37%,半乳糖差异不显著。结论:开花显著消耗铁皮石斛药材中的多糖及多糖中甘露糖、半乳糖醛酸、葡萄糖的量,而开花过程可能有利于木糖、阿拉伯糖积累。
Objective: To reveal the effect of flowering on the polysaccharide mass fraction and monosaccharide composition of Dendrobium candidum and provide the basis for the cultivation of Dendrobium officinale. Methods: Pre-flowering samples were collected from Dendrobium officinale. Samples of buds and buds were removed by hand before flowering. The content of polysaccharides in D. officinale was determined by phenol-sulfuric acid method. Results: The absolute amount of mannose, galacturonic acid and glucose in total polysaccharides and polysaccharides were 30.35%, 26.44%, 20.78% and 57.80% 28.33%, 24.37%, galactose difference was not significant. Conclusion: Flowering significantly consumes the amount of mannose, galacturonic acid and glucose in the polysaccharides and polysaccharides of Dendrobium officinale, and the flowering process may be beneficial to the accumulation of xylose and arabinose.
