目的:观察蓝莓对大鼠免疫性肝纤维化超微结构及细胞色素P4502E1(CYP2E1)表达的影响。方法:50只雄性Wistar大鼠随机分为正常组、模型组、蓝莓原浆预防组(蓝莓组)、复方鳖甲软肝片预防组(复方组)、蓝莓原浆加复方鳖甲软肝片预防组(蓝莓+复方组)。猪血清复制大鼠肝纤维化模型,各预防组在造模同时给予蓝莓原浆或(和)复方鳖甲软肝片灌胃。12周后处死大鼠,观察肝组织超微结构变化,RT-PCR、Western Blot和免疫组织化学法检测大鼠肝组织CYP2E1 mRNA及蛋白质的表达。结果:与模型组比,3个用药组肝纤维化程度明显减轻(P<0.05),超微结构病变明显改善,肝组织CYP2E1 mRNA及蛋白质表达无明显差异。结论:蓝莓对大鼠免疫性肝纤维化有一定的预防作用,对肝细胞超微结构病变有一定的保护作用,对CYP2E1的表达无明显影响。
Objective: To observe the effect of blueberry on the ultrastructure of immunological liver fibrosis and the expression of cytochrome P4502E1 (CYP2E1) in rats. Methods: Fifty male Wistar rats were randomly divided into normal group, model group, blueberry puree prevention group (blueberry group), Fufang Biejia Ruangan Pian preventing group (compound group), blueberry puree plus Fufang Biejia Ruangan Pian Prevention group (blueberry + compound group). Pig serum replication rat liver fibrosis model, the prevention group in the same time giving blueberry puree or (and) Fufang Biejia Ruangan tablets gavage. After 12 weeks, the rats were sacrificed and the ultrastructural changes of liver tissue were observed. The expression of CYP2E1 mRNA and protein in liver tissue were detected by RT-PCR, Western Blot and immunohistochemistry. Results: Compared with the model group, the degree of hepatic fibrosis in the three treatment groups was significantly reduced (P <0.05), the ultrastructure lesions were significantly improved, and there was no significant difference in the expression of CYP2E1 mRNA and protein in the liver tissue. CONCLUSION: Blueberry has some preventive effects on immune hepatic fibrosis in rats, and has certain protective effect on the ultrastructural changes of hepatocytes, but has no obvious effect on the expression of CYP2E1.
