目的 研究自身免疫病人外周血TCRγδ细胞系在微生物及寄生虫抗原刺激下细胞因子的分泌情况。方法 以TCRγδ单克隆抗体固相法体外选择性扩增获得7例胰岛素依赖型糖尿病(IDDM)及3例类风湿性关节炎(RA)病人外周血TCRγδ细胞系,平均纯度可达(847±59)%。在体外培养条件下,以植物血凝素(PHA)、结核分枝杆菌(M.tb)、阴道毛滴虫(TV)、人类6型疱疹病毒(HHV6)刺激24h及48h后,测定细胞因子分泌情况。结果 HHV6刺激组IL2的分泌量显著高于其它各实验组;M.tb及HHV6的刺激可导致IFNγ的高产量。TNFα的产生未见有显著性差异。结论 γδT细胞可能通过细胞因子的产生来影响自身免疫病的发生与发展。
Objective To study the secretion of cytokines by TCRγδ cells in peripheral blood of patients with autoimmune diseases stimulated by microorganisms and parasites. Methods Peripheral blood TCRγδ cell lines were obtained from 7 patients with insulin dependent diabetes mellitus (IDDM) and 3 patients with rheumatoid arthritis (RA) by selective amplification with TCRγδ monoclonal antibody. The average purity was (847 ± 59) )%. In vitro culture conditions, PHA, M. tuberculosis, Trichomonas vaginalis (TV), human herpesvirus 6 (HHV6) stimulation 24h and 48h after the determination of cytokines Secretion. Results The secretion of IL2 in HHV6-stimulated group was significantly higher than that in other experimental groups. The stimulation of M.tb and HHV6 resulted in high IFNγ production. There was no significant difference in TNFα production. Conclusion γδT cells may affect the occurrence and development of autoimmune diseases through the production of cytokines.
