目的比较凝血酶对培养的人血管内皮细胞(VEC)和U937细胞表达组织因子(TF)和尿激酶型纤溶酶原激活剂受体(uPAR)mRNA的影响,并探讨其机制。方法在凝血酶、无活性的凝血酶(DFP-T)和(或)抗凝血酶受体(TR)14肽多抗作用下,以RT-PCR法测定培养细胞TF和uPARmRNA,进行分析比较。结果TR抗体能完全阻断凝血酶诱导U937细胞表达TF和uPARmRNA;对VEC的作用并不完全。DFP-T对U937细胞无诱导作用;对VEC有低水平的诱导作用。结论凝血酶诱导TF和uPARmRNA表达的机制主要与其和细胞膜上TR结合并激活TR有关,在VEC可能存在TR以外的其他作用途径。
Objective To compare the effects of thrombin on the expression of tissue factor (TF) and urokinase-type plasminogen activator receptor (uPAR) mRNA in cultured human vascular endothelial cells (VEC) and U937 cells and to explore its mechanism. Methods The TF and uPAR mRNA in cultured cells were assayed by RT-PCR with polyclonal antibody against thrombin, inactive thrombin (DFP-T) and / or antithrombin receptor (TR) 14 . Results TR antibody could completely block the expression of TF and uPAR mRNA induced by thrombin in U937 cells; the effect on VEC was not complete. DFP-T has no effect on U937 cells and low level of VEC. Conclusions The mechanism of thrombin-induced TF and uPAR mRNA expression is mainly related to the binding of TR on the cell membrane and the activation of TR. There may be other pathways other than TR in VEC.
