目的:探讨有关层粘连蛋白受体(LNR)、上皮性钙调蛋白(E-cad)及Ⅳ型胶原酶(Ⅳcol)在大肠癌浸润中的相互关系。方法:利用人羊膜浸润模型,观察LNR、E-cad对大肠癌细胞株与羊膜粘附及细胞内Ⅳcol表达的影响。结果:高转移细胞株LoVo与羊膜的粘附率高于转移力较低细胞株HT29。瘤细胞用E-cad单抗封闭后培养于羊膜上,两细胞株内Ⅳ型胶原酶表达明显增高;但用LNR单抗封闭细胞后,细胞内Ⅳ型胶原酶表达降低,并且均以高转移株变化更为明显。结论:E-cad与LNR通过细胞内信使传递作用影响细胞内Ⅳ型胶原酶的表达,从而对肿瘤浸润和转移产生影响。
Objective: To investigate the relationship between laminin receptor (LNR), epiregulin calming (E-cad) and type IV collagenase (IVcol) in the invasion of colorectal cancer. METHODS: Using human amniotic membrane infiltration model, the effects of LNR and E-cad on the adhesion of colorectal cancer cell line and amniotic membrane and the expression of intracellular IVcol were observed. RESULTS: The adhesion rate of the highly metastatic cell line LoVo to the amniotic membrane was higher than that of the lower metastatic cell line HT29. The tumor cells were blocked with E-cadma and then cultured on the amniotic membrane. The expression of type IV collagenase in the two cell lines was significantly increased. However, when the cells were blocked with LNR, the expression of type IV collagenase in the cells was reduced, and all of them were highly metastatic. Changes in the strain are even more pronounced. CONCLUSION: E-cadherin and LNR affect the expression of type IV collagenase in cells through intracellular messenger transmission, and thus have effects on tumor invasion and metastasis.
