目的探讨通络救脑注射液及其有效成分对脑微血管内皮细胞的保护作用及机制。方法建立β淀粉样蛋白(Aβ_(1-40))致人脑微血管内皮细胞(h BMECs)损伤模型,用通络救脑注射液及其有效单体成分京尼平苷、人参皂苷Rg1干预,采用CCK-8染色法测定细胞活性,检测条件培养液中乳酸脱氢酶(LDH)漏出量,Western blot法检测h BMECs高度糖基化终产物受体(RAGE)的表达。结果通络救脑注射液及其有效单体成分京尼平苷、人参皂苷Rg1可显著提高Aβ_(1-40)损伤的h BMECs活性,减少细胞损伤后LDH的漏出和h BMECs RAGE的表达,与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论通络救脑注射液及其有效单体成分京尼平苷、人参皂苷Rg1可提高Aβ_(1-40)损伤的h BMECs的活性,对其具有保护作用,其机制可能与减少血管内皮细胞RAGE的表达有关。
Objective To investigate the protective effect of Tongluo Jiu Nao injection and its active ingredients on brain microvascular endothelial cells and its mechanism. Methods The injury models of human BMMCs induced by β-amyloid protein (Aβ 1-40) were established. Intervention with Tongluo Jiuna injection and its effective monomer components geniposide, ginsenoside Rg1, Cell viability was measured by CCK-8 staining, lactate dehydrogenase (LDH) leakage in conditioned medium was detected, and the expression of highly glycated end product receptor (RAGE) in h BMECs was detected by Western blot. Results Tongluo Jianyu injection and its effective monomer component of geniposide and ginsenoside Rg1 could significantly increase the activity of h BMECs induced by Aβ 1-40 and reduce the leakage of LDH and the expression of RAGE in h BMECs after cell injury, Compared with the model group, the differences were statistically significant (P <0.05, P <0.01). Conclusion Tonglianjinao injection and its effective monomer component of geniposide and ginsenoside Rg1 can enhance the activity of hβ BMECs injured by Aβ 1-40, which may be related to the decrease of vascular endothelial cells RAGE expression.
