目的:观察降压通络方对高血压肾损害大鼠肾脏及血浆肾上腺髓质素(adrenomedullin,ADM)表达的影响,探讨降压通络方对高血压肾损害大鼠肾脏的保护机制。方法:将16周龄自发性高血压大鼠(spontaneously hypertension rat,SHR)随机分为模型组、缬沙坦组、降压通络方组,同时设16周龄正常血压大鼠(wistar kyoto,WKY)为正常对照组。缬沙坦组、降压通络方组分别给予缬沙坦、降压通络方灌胃干预,模型组及正常对照组大鼠给予等容积的生理盐水灌胃,连续8周。分别于给药前、给药4周、给药8周末测定大鼠血压;给药4周、8周末,检测血肌酐、尿素氮及肾脏、血浆ADM的含量。结果:给药4周、8周后,降压通络方组、缬沙坦组大鼠血压、肌酐、尿素氮均低于模型组(P<0.05或P<0.01);而大鼠肾脏ADM含量高于模型组(P<0.05或P<0.01);给药4周后,模型组及降压通络方组、缬沙坦组大鼠血浆ADM明显高于正常对照组,8周后模型及降压通络方组、缬沙坦组血浆ADM低于正常对照组,但两个时间点模型组与降压通络方组、缬沙坦组之间比较,差异无统计学意义。结论:降压通络方可降低高血压肾损害大鼠血压,保护肾功能,可能机制为上调肾脏局部ADM的表达。
Objective: To observe the effect of Jiasha Tongluo Recipe on the expression of renal and plasma adrenomedullin (ADM) in hypertensive renal tissue in rats and to explore the protective mechanism of Jiasha Tongluo recipe on the kidney of hypertensive renal injured rats. Methods: 16-week-old spontaneously hypertensive rats (SHR) were randomly divided into model group, valsartan group and JSTL group. At the same time, 16-week-old normal rats (wistar kyoto, WKY) as a normal control group. Valsartan group, JSTT group were given valsartan, antihypertension Tongluo prescription gavage intervention, model group and normal control group rats were given equal volume of saline for 8 weeks. The blood pressure of rat was measured before administration, 4 weeks after administration and 8 weeks after administration. Serum creatinine, urea nitrogen, renal and plasma ADM were measured at the end of 4 weeks and 8 weeks after administration. Results: The blood pressure, creatinine and urea nitrogen in JJL and Valsartan groups were lower than those in model group (P <0.05 or P <0.01) (P <0.05 or P <0.01). After 4 weeks of administration, the levels of plasma ADM in model group, JJT group and Valsartan group were significantly higher than those in normal control group. After 8 weeks, And lowering blood pressure and relieving collaterals group and valsartan group plasma ADM was lower than the normal control group, but the two time points model group and the Jiasha Tongluo prescription group, valsartan group comparison between the difference was not statistically significant. Conclusion: JJT can reduce blood pressure and protect renal function in rats with hypertensive renal failure. The possible mechanism is to increase the expression of local ADM in the kidney.
