目的研究姜黄挥发油(TVO)对急性单核细胞白血病THP-1细胞株增殖及凋亡的影响,并探讨其作用机制。方法不同浓度(5~80 mg/L)TVO体外作用于THP-1细胞。应用噻唑蓝(MTT)法检测细胞增殖抑制率;经吉姆萨染色,倒置显微镜观察细胞形态变化;DNA片段化测细胞凋亡;流式细胞仪检测细胞凋亡及周期;比色法测Caspase-3酶活性。结果 TVO具有抑制THP-1细胞株增殖的作用,并表现出时间-剂量依赖性。同时,TVO能诱导THP-1细胞凋亡,呈现剂量依赖性,镜下见典型细胞凋亡形态,Caspase-3酶活性随药物浓度增加而增强。经TVO作用后的细胞周期被阻滞在G1期,G2/M期及S期细胞数减少。结论 TVO对THP-1细胞株具有增殖抑制及促凋亡的作用,其机制可能是使细胞周期阻滞在G1期,且激活了细胞凋亡途径中关键酶Caspase-3活性,从而抑制肿瘤细胞的增殖与分化。
Objective To study the effect of volatile oil of turmeric (TVO) on the proliferation and apoptosis of acute monocytic leukemia THP-1 cell line and to explore its mechanism. Methods Different concentrations of TVO (5 ~ 80 mg / L) were administered to THP-1 cells in vitro. The cell proliferation inhibition rate was detected by MTT assay; the morphological changes of cells were observed by Giemsa staining; the apoptosis of cells was detected by DNA fragmentation; the apoptosis and cycle were detected by flow cytometry; the expressions of Caspase- 3 enzyme activity. Results TVO could inhibit the proliferation of THP-1 cell line and showed time-dose dependence. At the same time, TVO could induce apoptosis of THP-1 cells in a dose-dependent manner. Typical apoptotic morphology of THP-1 cells was observed under microscope. Caspase-3 activity increased with the increase of drug concentration. After the TVO effect cell cycle was blocked in the G1 phase, G2 / M phase and S phase cells decreased. Conclusion TVO can inhibit proliferation and induce apoptosis of THP-1 cell line by blocking cell cycle in G1 phase and activating caspase-3, a key enzyme in apoptosis pathway, thereby inhibiting tumor cells Proliferation and differentiation.
