本研究比较了5种方法从棉花叶片中提取棉花曲叶病毒(CLCuV)基因组DNA的提取效果。对不同的CLCuV分离物的PCR扩增表明,通常用于双生病毒基因组DNA提取的NaOH法和CTAB提取的DNA无法用于PCR检测,但CTAB法提取的DNA可用于Southernblot杂交。有3种方法则可有效地提取CLCuVDNA,供PCR扩增使用,这3种方法的共同点是去除了棉花组织中的干扰物质。
In this study, five methods were compared to extract cotton leaf curl cotton (CLCuV) genomic DNA extraction. PCR amplification of different CLCuV isolates showed that the NaOH and CTAB DNAs commonly used for genomic DNA extraction from gemini can not be used for PCR, but DNA extracted by CTAB can be used for Southernblot hybridization. There are three ways to efficiently extract CLCuVDNA for PCR amplification. The three methods have in common the removal of interfering substances in cotton tissue.
