目的:探讨海带多糖(LJP)对放疗后颌下腺血管的影响。方法:70只Wistar大鼠随机分为正常对照组(N),放疗对照组(R),海带多糖高剂量(300mg/kg)+照射组(LRA),海带多糖低剂量(30mg/kg)+照射组(LRB)。照射前后3d分别给予腹腔注射0.9%氯化钠溶液和海带多糖。R组,LRA组,LRB组的大鼠头颈部予以60Coγ射线15Gy一次性照射。透射电镜观察颌下腺血管内皮细胞超微结构改变,免疫组化法检测腺体微血管密度(MVD),RT-PCR检测血管内皮生长因子(VEGF),基质金属蛋白酶9(MMP-9)及其抑制剂(TIMP-1)的表达。结果:电镜观察LRA及LRB组血管内皮细胞损伤较轻;免疫组化结果显示,与R组相比,LRA及LRB组MVD较高(P<0.05);RT-PCR检测VEGF、MMP-9在LRA及LRB组的表达高于R组(P<0.05),TIMP-1在各组表达无差异。结论:LJP可减轻射线对颌下腺血管的损伤。
Objective: To investigate the effect of laminarin (LJP) on the submandibular gland vessels after radiotherapy. Methods: Seventy Wistar rats were randomly divided into normal control group (N), radiotherapy control group (R), high dose of Laminar (300mg / kg) + irradiation group, low dose of Laminar (30mg / kg) Irradiation group (LRB). Intraperitoneal injection of 0.9% sodium chloride solution and kelp polysaccharide were given before and after irradiation respectively. R group, LRA group, LRB group of rats head and neck to 60Coγ ray 15Gy a one-time irradiation. The changes of ultrastructure of submandibular gland vascular endothelial cells were observed by transmission electron microscopy. The gland microvessel density (MVD) was detected by immunohistochemistry. The expressions of vascular endothelial growth factor (VEGF), matrix metalloproteinase 9 (MMP-9) and its inhibitor (TIMP-1) expression. Results: The damage of vascular endothelial cells in LRA and LRB groups was lesser by electron microscopy. The immunohistochemistry results showed that MVD in LRA and LRB groups was higher than that in R group (P <0.05) The expression of LRA and LRB was higher than that of R (P <0.05), while there was no difference of TIMP-1 in all groups. Conclusion: LJP can reduce the damage of rays to the submandibular gland.
