目的研究金丝桃苷联合长春新碱对人结肠癌HCT8细胞增殖和凋亡的影响,探讨其协同增效作用及机制。方法采用噻唑蓝(MTT)法检测不同浓度的金丝桃苷对HCT8细胞的增殖抑制作用,并计算IC20,IC50;采用IC20浓度的金丝桃苷联合不同浓度的长春新碱,观察对HCT8细胞增殖的影响;金丝桃苷联合长春新碱处理HCT8细胞,流式细胞仪检测细胞凋亡率,比色法检测Caspase-3和Caspase-9活性的变化。结果金丝桃苷能有效抑制HCT8细胞的增殖,IC20约为5mg/L,IC50约为30mg/L。金丝桃苷与长春新碱联合作用于HCT8细胞48h,可提高HCT8细胞对长春新碱的敏感性,增敏4.88倍。金丝桃苷与长春新碱联合用药细胞凋亡率、Caspase-3和Caspase-9活性比单独用金丝桃苷和长春新碱都有所提高(P<0.01)。结论金丝桃苷能提高人结肠癌HCT8细胞对长春新碱的敏感性,金丝桃苷联合长春新碱后会增强Caspase-3和Caspase-9活性,从而促进HCT8细胞凋亡。
Objective To study the effect of hyperoside combined with vincristine on proliferation and apoptosis of human colon cancer HCT8 cells and to explore its synergistic effect and mechanism. Methods MTT assay was used to detect the inhibitory effect of different concentrations of hyperoside on the proliferation of HCT8 cells, and the IC50 and IC50 were calculated. Using the hyperin with different concentration of IC20 and vincristine, HCT8 cells The proliferation of HCT8 cells was detected by hyperin and vinblastine. Flow cytometry was used to detect the apoptosis rate. Caspase-3 and Caspase-9 activity were detected by colorimetric assay. Results Hyperin effectively inhibited the proliferation of HCT8 cells with IC50 of about 5 mg / L and IC50 of about 30 mg / L. Hyperoside combined with vincristine on HCT8 cells 48h, HCT8 cells can increase the sensitivity of vincristine, sensitized 4.88 times. Hypericumin combined with vincristine apoptosis rate, Caspase-3 and Caspase-9 activity than the hyperoside alone and vincristine were increased (P <0.01). Conclusion Hyperin can increase the sensitivity of human colon carcinoma HCT8 cells to vincristine. Hyperin combined with vincristine can enhance the activity of Caspase-3 and Caspase-9 and promote the apoptosis of HCT8 cells.
