目的建立两种简便的白色念珠菌DNA提取方法,即纯DNA提取法和快速DNA提取法。方法以原生质体提取法为基础进行改良,两种方法均用Lyticase破壁。结果与结论纯NDA提取法提取DNA耗时6~7h,产量较高,每10~(10)个菌细胞可提取4μgDNA,且纯度高,OD_(260)OD_(280)>1.7,对核酸限制性内切酶敏感,适用于各种分子生物学方面的研究。快速DNA提取法全过程仅需3~4h,每10~(10)个菌细胞提取1μgDNA,OD_(260):OD_(280)=1.4,不能进行核酸限制性内切酶酶切,但可用于作PCR扩增。
Objective To establish two simple DNA extraction methods of Candida albicans, namely pure DNA extraction and rapid DNA extraction. The method was modified on the basis of protoplast extraction, both of which were broken with Lyticase. RESULTS AND CONCLUSION Pure DNA extraction (NDA) method took 6 ~ 7 hours to extract DNA, yielding 4μg DNA per 10 ~ 10 cells with high purity and OD 260 (280)> 1.7 Nucleic acid restriction endonuclease sensitive, suitable for a variety of molecular biology research. In the whole process of rapid DNA extraction, it takes only 3 ~ 4h to extract 1μg DNA per 10 ~ 10 bacterial cells.
