目的 研究3,4,5,6-四羟基(口山)酮对高糖诱导的血管内皮细胞损伤的保护作用。方法 在培养的人脐静脉内皮细胞(ECV304)高浓度葡萄糖(30mmol·L~(-1))处理48h引起细胞损伤。荧光分光光度法检测细胞培养液中乳酸脱氢酶(LDH)活性、一氧化氮(NO)生成量与细胞脂质过氧化物丙二醛(MDA)含量;MTT比色法检测细胞存活数量;流式细胞仪检测细胞周期。结果 ①3,4,5,6-四羟基(口山)酮能显著抑制高糖所致内皮细胞LDH释放、NO和MDA生成增加;②3,4,5,6-四羟基(口山)酮能显著抑制高糖所致的活细胞数减少;③3,4,5,6-四羟基(口山)酮能显著减轻高糖所致肉皮细胞增殖的抑制。结论 3,4,5,6-四羟基咄酮减轻高糖所致的人脐静脉内皮细胞损伤。其保护作用与抗氧化减少氧自由基生成有关。
Objective To study the protective effect of 3,4,5,6-tetrahydroxy-keto-ketone on the injury of vascular endothelial cells induced by high glucose. Methods Cell injury was induced by high concentration of glucose (30 mmol·L -1) in cultured human umbilical vein endothelial cells (ECV304) for 48 hours. Fluorescence spectrophotometry was used to detect lactate dehydrogenase (LDH) activity, nitric oxide (NO) production and lipid peroxidation (MDA) content in cell culture medium. Cell viability was measured by MTT assay. Flow Cytometry to detect cell cycle. Results ① 3, 4, 5, 6-tetrahydroxysteroid could significantly inhibit the release of LDH from endothelial cells induced by high glucose, and increase the production of NO and MDA; Significantly inhibited the number of viable cells induced by high glucose decreased; ③ 3,4,5,6-tetrahydroxyl (Kushan) ketone can significantly reduce the inhibition of high glucose-induced proliferation of human skin cells. Conclusion 3,4,5,6-tetrahydroxytrendione can reduce the damage of human umbilical vein endothelial cells induced by high glucose. Its protective effect and anti-oxidation reduce the generation of oxygen free radicals.
