目的 :了解抑癌基因 P5 3、癌基因 C- erb B- 2和增殖细胞核抗原 (PCNA )在子宫内膜增生过长和子宫内膜癌中的存在状况及意义。方法 :用 P5 3、C- erb B- 2和 PCNA抗体对 5 0例子宫内膜癌和 2 0例子宫内膜增生过长手术标本进行 S- P免疫组化检测 ,分析表达率与病理分级的关系。结果 :P5 3、C- erb B- 2和 PCNA在子宫内膜癌组中的表达分别为 5 4%、 5 6 %和72 % ,均高于子宫内膜增生过长组。P5 3、C- erb B- 2和 PCNA均随内膜癌的恶性程度增高而呈增高趋势 ,P5 3和 PCNA的 级表达明显高于 级 (P <0 .0 1)。研究结果提示 :子宫内膜癌的发生与 P5 3和 C- erb B- 2基因蛋白的过度表达或突变及 PCNA的过度表达有关 ;P5 3、C- erb B- 2和 PCNA之间可能有相互协同作用 ;P5 3、C- erb B- 2和 PCNA表达反映了瘤细胞的分化程度及增殖情况 ,因而可作为子宫内膜癌的诊断、鉴别诊断及预后的参考指标之一。
Objective: To investigate the status and significance of tumor suppressor gene P53, C-erb B- 2 and proliferating cell nuclear antigen (PCNA) in endometrial hyperplasia and endometrial carcinoma. Methods: Fifty cases of endometrial carcinoma and 20 cases of endometrial hyperplasia surgically treated with P5 3, C-erb B-2 and PCNA antibody were detected by S-P immunohistochemistry. The expression and pathological grade Relationship. Results: The expressions of P53, C-erb B-2 and PCNA in endometrial carcinoma were 54%, 56% and 72%, respectively, which were higher than those in endometrial hyperplasia group. The expression of P53, C-erb B-2 and PCNA both increased along with the malignant degree of endometrial carcinoma, and the expression of P53 and PCNA was significantly higher than that of grade (P <0.01). The results suggest that: the occurrence of endometrial cancer P5 3 and C-erb B-2 gene protein overexpression or mutation and PCNA over-expression; P5 3, C-erb B-2 and PCNA may have mutual Synergy; P5 3, C-erb B-2 and PCNA expression reflects the degree of tumor cell differentiation and proliferation, which can be used as endometrial cancer diagnosis, differential diagnosis and prognosis of the reference index.
