目的探讨成年大鼠局灶性脑缺血后齿状回颗粒下层(SGZ)神经元前体细胞的增殖改变。方法大脑中动脉栓塞再灌注(MCAO/R),术后第1天、第2天腹腔注射Brdu,第3天、7天、14天和28天取脑,每个时间组6只SD大鼠,冷冻切片,免疫荧光三标检测SGZ区5-溴脱氧尿嘧啶核苷(Brdu)、DCX、ki67、胶质纤维酸性蛋白(GFAP)及CNP的表达。结果 (1)MCAO/R后,SGZ区增殖细胞聚集成簇,细胞簇计数从3d开始增加,7d时最多,随后下降;(2)细胞簇内细胞计数从3d开始增加,7d时最多,随后下降;(3)细胞簇中ki67(+)细胞比例从3d开始到28d逐渐下降。结论 MCAO/R引发SGZ区神经元前体细胞经多次分裂以成簇方式增殖,部分子代细胞向神经元分化,部分子代细胞仍保持增殖能力。
Objective To investigate the proliferation of adult rat SGZ neurons after focal cerebral ischemia. Methods Middle cerebral artery occlusion and reperfusion (MCAO / R) were performed. Brdu was intraperitoneally injected on the first day and the second day after operation. Brain was taken on the 3rd, 7th, 14th and 28th day. Six SD rats , Frozen section and immunofluorescence were used to detect the expression of Brdu, DCX, ki67, glial fibrillary acidic protein (GFAP) and CNP in SGZ. Results (1) After MCAO / R, proliferating cells clustered into clusters in SGZ, the cell cluster counts began to increase from 3d, and reached the maximum at 7d, then decreased; (2) The intracellular cluster cell counts increased from 3d to 7d, (3) The proportion of ki67 (+) cells in the cluster gradually decreased from 3d to 28d. Conclusion MCAO / R induced neuronal progenitor cells in SGZ area to proliferate in multiple ways through multiple division. Some progeny differentiate into neurons, and some progeny cells still maintain their proliferative capacity.
