目的:建立HPLC测定芎菊上清颗粒中绿原酸、栀子苷、盐酸小檗碱和黄芩苷4种成分含量的方法。方法:色谱柱为DIKMA Platisil ODS色谱柱(250 mm×4.6 mm,5μm),柱温30℃,流动相为甲醇-0.1%磷酸水溶液,采用梯度洗脱,检测波长为240 nm,流速为1.0 mL/min。结果:绿原酸、栀子苷、盐酸小檗碱和黄芩苷的线性范围分别为0.01~0.2μg(r1=0.9998)、0.1~2μg(r2=0.9997)、0.02~0.4μg(r3=0.9998)、0.017~0.34μg(r4=0.9999),平均回收率为98.51%~99.13%。结论:该法检测效率高、专属性强、重复性好,可用于芎菊上清颗粒中绿原酸、栀子苷、盐酸小檗碱和黄芩苷的定量测定。
OBJECTIVE: To establish a HPLC method for the determination of the contents of 4 components in chlorogenic acid, geniposide, berberine and baicalin in the supernatant granules of Chuan Ju. METHODS: The column was a DIKMA Platisil ODS column (250 mm × 4.6 mm, 5 μm) with a mobile phase of methanol-0.1% phosphoric acid at a temperature of 30 ℃. The gradient elution was carried out at a detection wavelength of 240 nm and a flow rate of 1.0 mL / min. Results: The linear ranges of chlorogenic acid, geniposide, berberine and baicalin were 0.01-0.2μg (r1 = 0.9998), 0.1-2μg (r2 = 0.9997), 0.02-0.4μg (r3 = 0.9998) , 0.017 ~ 0.34μg (r4 = 0.9999), the average recovery was 98.51% ~ 99.13%. Conclusion: The method has high detection efficiency, specificity and repeatability, and can be used for the quantitative determination of chlorogenic acid, geniposide, berberine hydrochloride and baicalin in the granules of Xuanju supernatant.
