以7株贵州蜜环菌为试材,采用苯酚-硫酸法测定多糖得率、DNS法测定蜜环菌发酵液中木聚糖酶和羧甲基纤维素酶的活性、丁香醛连氮测定发酵液中漆酶活性,研究了不同蜜环菌在发酵过程中漆酶等胞外酶活性变化规律及菌丝胞内多糖提取率。结果表明:各菌株间多糖得率存在显著差异;各菌株胞外酶活性变化趋势均是先升后降;各酶在不同菌株间达到最大酶活的时间、最大酶活性值存在明显差异;优良菌株DJ1第4天就能结束发酵,达到最大酶活时间最短、持续时间长。蜜环菌液体发酵时最早分泌的是CMC酶、漆酶,然后是木聚糖酶,但起主要作用的是木聚糖酶。
Seven strains of Armillaria mellea in Guizhou were used as materials to determine the yield of polysaccharides by phenol-sulfuric acid method. The activity of xylanase and carboxymethylcellulase in Armillaria mellea was determined by DNS method. Liquid laccase activity of different Armillaria mellea bacteria in the fermentation process of laccase and other extracellular enzyme activity changes and mycelium intracellular polysaccharide extraction rate. The results showed that there was a significant difference in the yield of polysaccharides between the strains. The trend of extracellular enzyme activity of all the strains increased first and then decreased. The maximum enzyme activity reached the maximum between different strains, and the maximum enzyme activity value was significantly different. Strain DJ1 can end fermentation on the 4th day to reach the maximum enzymatic activity for the shortest duration. Meloidogyne liquid fermentation is the earliest secretion of CMC enzyme, laccase, followed by xylanase, but played a major role in xylanase.
