目的观察维生素E对锰诱导的PC12细胞毒性的抑制效应,从而为锰神经毒性的防护提供可能的途径。方法 MTT、TUNEL、姬姆萨染色、流式细胞仪检测细胞毒性;试剂盒检测MDA含量以及T-SOD、MnSOD活性的变化。结果锰可以诱导PC12细胞氧化应激水平显著增高,显示为MDA含量的增高和T-SOD、MnSOD活性代偿性升高。维生素E可以抑制锰诱导的PC12细胞毒性,表现为其可抑制锰诱导的细胞活力的下降以及凋亡率的升高。结论维生素E作为抗氧化剂可以抑制由锰诱导的PC12细胞的一系列细胞毒性,补充维生素可作为一种防护锰毒性的有效手段。
Objective To observe the inhibitory effect of vitamin E on the manganese-induced cytotoxicity of PC12 cells, and to provide a possible way to protect the neurotoxicity of manganese. Methods MTT, TUNEL, Giemsa staining, cytotoxicity were detected by flow cytometry. The contents of MDA and the activities of T-SOD and MnSOD were detected by ELISA kit. Results Manganese induced a marked increase of oxidative stress in PC12 cells, indicating the increase of MDA content and the compensatory increase of T-SOD and MnSOD activities. Vitamin E inhibits manganese-induced PC12 cytotoxicity by showing that it inhibits manganese-induced decrease of cell viability and increase of apoptosis rate. Conclusion Vitamin E as an antioxidant can inhibit a series of cytotoxicity induced by manganese in PC12 cells. Vitamin supplementation can be used as an effective measure to protect against manganese toxicity.
