电镜诊断学是指对临床某些疑难病例,应用综合电镜技术,列出标本制备常规流程,确定诊断指标及鉴别诊断指标,在超微结构水平上作出明确诊断的一门新兴应用方法学。我们自70年代初开始,对白血病进行了系列的电镜观察,建立了适用于白血病诊断的综合电镜技术,确定了可靠的诊断指标。经1000多例次的电镜诊断和临床疗效验证,其确诊率由原来用光镜分类的60~70%(按FAB分类)提高到99%以上。该方法学主要内容包括:白血病超微结构形态分类、酶细胞化学诊断、免疫细胞化学分型诊断、电镜标本制作常规流程。
Electron microscopy diagnostics refers to the application of comprehensive electron microscopy to certain clinically difficult cases, listing routine routines for specimen preparation, determining diagnostic criteria and differential diagnostic indicators, and establishing an emerging diagnostic methodology for definitive diagnosis at the ultrastructural level. Since the early 1970s, we have conducted a series of electron microscopy observations on leukemia, established a comprehensive electron microscopy technique for the diagnosis of leukemia, and established reliable diagnostic indicators. After more than 1,000 cases of electron microscopy diagnosis and clinical efficacy verification, the final diagnosis rate was increased from 60 to 70% (by FAB classification) with light microscope classification to more than 99%. The main contents of the methodology include: ultrastructural morphological classification of leukemia, enzyme cytochemical diagnosis, immunocytochemical type diagnosis, and routine procedures for electron microscopic specimen preparation.
