目的 :探讨人补体C5b~ 9复合物对大鼠肾小球系膜细胞 (MC)合成一氧化氮 (NO)的影响。方法 :首先提取人补体C5b~ 9复合物 ,同时培养出大鼠肾小球MC。然后用C5b~ 9复合物刺激MC ,观察刺激后 6h、2 4h、和 48h培养上清液中NO代谢产物 -硝酸根 (NO-3 )和亚硝酸根 (NO 2 )量的变化 ,并测定刺激后 2 4h、48h时MC内环鸟苷酸 (cGMP)的水平。结果 :用人C5b~ 9复合物刺激大鼠MC后可致培养上清中NO 3 /NO 2 含量增加 ,细胞内cGMP水平上升。NO合成增多能够被NO合酶 (NOS)抑制剂—─ -NG-硝基 -L -精氨酸甲基酯 (L -NAME)所抑制。结论 :人C5b~ 9复合物能够增加大鼠肾MC的NO合成。
Objective: To investigate the effect of human complement C5b ~ 9 complex on the synthesis of nitric oxide (NO) in rat mesangial cells (MC). Methods: Firstly, human complement C5b ~ 9 complex was extracted and glomerular MC was cultured at the same time. Then, the MCs were stimulated with C5b ~ 9 complex and the changes of NO metabolites such as NO-3 and NO2 in supernatants of 6h, 24h and 48h after stimulation were observed. After 24 h, 48 h, the level of cGMP in MCs was increased. Results: Stimulation of rat MC with C5b ~ 9 complex resulted in an increase of NO3 / NO2 content in the culture supernatant and an increase of intracellular cGMP levels. Increased NO synthesis can be inhibited by the NO synthase (NOS) inhibitor, -NG-nitro-L-arginine methyl ester (L -NAME). Conclusion: Human C5b ~ 9 complex can increase NO synthesis in rat kidney MC.
