目的探讨二甲双胍和顺铂联合用药对B16F10黑素瘤细胞的影响。方法体外培养黑素瘤细胞株B16F10,并用一定浓度的二甲双胍和顺铂处理肿瘤细胞。采用MTT法检测肿瘤细胞活性,并用流式细胞术检测细胞周期和凋亡情况。结果 MTT结果表明,二甲双胍顺铂联用组细胞活性显著低于药物单用组(P<0.05),二甲双胍顺铂存在对肿瘤的协同抑制作用(CDI<1);流式细胞术结果表明,二甲双胍顺铂联用组的凋亡细胞比率(23.21±4.11)%明显高于空白对照组(1.21±0.12)%、二甲双胍组(2.32±0.16)%及顺铂组(14.12±1.78)%(P<0.05)。结论二甲双胍与顺铂具有协同抑制黑素瘤生长的作用,并且和凋亡诱导效应的增强相关。
Objective To investigate the effect of combination of metformin and cisplatin on B16F10 melanoma cells. Methods The melanoma cell line B16F10 was cultured in vitro and the tumor cells were treated with metformin and cisplatin at a certain concentration. The activity of tumor cells was detected by MTT assay, and cell cycle and apoptosis were detected by flow cytometry. Results The results of MTT showed that the cell viability of metformin combined with cisplatin was significantly lower than that of the drug alone group (P <0.05), and the synergistic inhibition of cisplatin on the tumor was found (CDI <1). The results of flow cytometry showed that metformin The percentage of apoptotic cells in cisplatin combination group was significantly higher than that in control group (23.21 ± 4.11)% (1.21 ± 0.12)%, metformin group (2.32 ± 0.16)% and cisplatin group (14.12 ± 1.78)% 0.05). Conclusion Metformin and cisplatin have a synergistic effect in inhibiting the growth of melanoma and are related to the enhancement of apoptosis-inducing effect.
