目的:了解免疫荧光层析法检测血清降钙素原(PCT)的性能。方法:用电化学发光法(ECLIA)和免疫荧光层析法平行检测300例血清样本PCT的浓度,(1)用Kappa检验分别评价Cut-off值为0.5ng/ml和2.0ng/ml时2种方法的一致性,Kappa值≥0.75时为吻合度好。(2)PCT>0.5ng/ml的样本200例,以ECLIA法为比对方法(X),以免疫荧光法为实验方法(Y),结果进行线性回归和相关系数(R)的计算。结果:(1)Cut-off值为0.50ng/ml和2.00ng/ml时,Kappa值分别为0.780和0.854,免疫荧光法和ECLIA法的吻合度好。(2)PCT>0.50ng/ml的样本,直线回归方程为Y=0.747X-0.158,R=0.985。结论:免疫荧光层析法检测血清PCT和ECLIA法检测的结果相关性高,吻合度好,适用于各类医院血清PCT的急诊检验。
Objective: To understand the performance of serum procalcitonin (PCT) by immunofluorescence assay. Methods: The concentrations of PCT in 300 serum samples were detected in parallel by ECLIA and immunofluorescence. CONCLUSION: The results of immunofluorescence assay for detection of serum PCT and ECLIA have high correlation and good agreement, and are suitable for the emergency examination of serum PCT in various hospitals.
