以新疆辣椒主栽品种“四平头”(带毒率较高)为试材,切取无菌苗生长点,以MS为基本培养基,在其上分别添加6-苄基腺嘌呤(6-BA)、吲哚乙酸(IAA)、玉米素(ZT)、细胞激动素(KT)、赤霉素(GA3)和萘乙酸(NAA)、苯乙酸(PAA)以及吲哚丁酸(IBA)等生长调节剂进行培养。经筛选80余种不同激素水平的培养基,其中以MS+BA8+IAA0.05~1.0较适宜茎尖的培养,MS+KT0.2+GA30.5~5或MS+ZT8+GA32~3较适宜芽的伸长。将伸长1cm以上的芽转入MS+IBA2或MS无激素的培养基上利于根的生长,生根后移入土壤即可长成正常植株。成苗脱毒率达100%。
